A closer look inside the sperm nucleus

The study, published in Nature Structural & Molecular Biology, challenges the long-held view that the sperm nucleus is simply a tightly compressed package of DNA. Instead, the researchers reported small DNA-free cavities that appear to serve as dedicated hubs for proteasomes. According to the team, these structures were observed with cryo-electron tomography, which allowed them to examine sperm cells in their native state at extremely high resolution.

The researchers said the cavities, which microscopists had noted years ago and called “lacunae,” had not previously been understood in biological terms. Their new analysis showed that the cavities contain large numbers of proteasomes arranged into distinct clusters, and that sperm proteasomes also carry unique features, including a previously unknown variant of a sperm-specific proteasome component.

Why the findings matter for fertility research

The team also studied human testicular tissue to learn when these structures form. They found that the proteasome-rich compartments emerge during the final stages of sperm development, when genetic material is being reorganized and compacted. That timing suggests the structures may play a role in sperm maturation and may also remain active after fertilization, potentially helping to remodel the paternal genome in the earliest stages of embryonic development.

Prof. Tom Dendooven of the VIB-VUB Center for Structural Biology said the team was surprised by the level of organization inside the sperm head and described the discovery as a new framework for studying how sperm cells develop and what happens immediately after fertilization. The researchers said their work provides the first detailed map of proteasome organization inside the human sperm nucleus and creates a foundation for future work on fertility, reproductive health and early embryonic development.

What comes next

While the findings do not by themselves explain infertility, they offer a new structural target for scientists investigating how sperm cells mature and how errors in that process could affect reproductive outcomes. The study adds to a growing body of research showing that even highly specialized human cells can contain unexpectedly complex internal machinery, and it could help guide future studies aimed at understanding male reproductive health more precisely.
